ABSTRACT: The secretion of type I procollagen presents a classic biophysical paradox: 300–400 nm triple-helical rods must traverse secretory pathways governed by 60–90 nm COPII vesicles. Overturning long-standing dogma, recent insights demonstrate that procollagen undergoes liquid-liquid phase separation within the endoplasmic reticulum (ER) lumen. This fluid state accommodates high cargo density while preventing premature intracellular aggregation. At ER exit sites, the master organizer Transport ANd Golgi Organization 1 (TANGO1) acts as a fluid-extrusion funnel, capturing these dynamic condensates and recruiting retrograde endoplasmic reticulum–Golgi intermediate compartment (ERGIC) membranes to form transient intercompartmental tunnels. As the condensate flows toward the Golgi, progressive changes in the luminal environment trigger a controlled liquid-to-crystalline phase transition that packs procollagen into dense, parallel arrays. Following exocytosis, extracellular metalloproteinases cleave the solubilizing N- and C-propeptides. Stripping the protective hydrophilic caps shifts the system toward hydrophobicity, driving entropy-driven self-assembly into striated fibrils that cross-linking then locks in place. Reframing procollagen export through fluid dynamics and colloidal phase transitions redefines basic organellar logistics. This review discusses the current literature on the fluid procollagen condensate in the ER and the TANGO1-mediated transient tunnel framework. It also raises some profound biochemical, structural, and physiological questions that remain.
